Hi Ben and Marina.
How long does it take to get to Plaza Major from Plaza Colon bus terminal?

Metro is the best option if you have lugagge. However you could just walk and still it will take about 30 minutes and you get some interesting sites in the way. Like Cafe Gijon, Palacio de Comunicaciones (Correos - The main post office in Madrid), Cibeles, Circulo de Bellas Artes and Sol, apart from Colon and Plaza Mayor.
I suppose you are coming from the airport, in that case just note that you can travel by metro directly from the airport if you arrive to terminals 1, 2 & 3.
But from terminal 4 either you take a bus to Barajas Airport metro Stations (apparently it runs very often and it only takes 5 minutes) or you take the bus to Colon.

The airport bus no longer runs to colon. I think it may still go to Avenida de America though...
